Initial Seeding Density:

0.6–1.0 × 10⁶ cells/mL. Seed cells should be in the logarithmic growth phase with a viability of no less than 95%.


Culture Conditions:

Temperature: 37°C
Humidity: 80% relative humidity
CO₂: 5%


Agitation:

120–150 rpm, 50mm (reduce speed appropriately for larger volumes).


Optional Passaging:

If conditions allow, pass cells once or more in the three basal media prior to inoculation. Alternatively, direct inoculation into the test media can be conducted, in which case centrifugation and resuspension is recommended to minimize the influence of previous media on screening results.
